beta Arrestin 1 Antibody (YA5236) is a Mouse-derived and non-conjugated monoclonal antibody, targeting to beta Arrestin 1.

Background

β-Arrestin 1 is a multifunctional regulatory protein primarily involved in modulating G-protein coupled receptor (GPCR) signaling, including receptor desensitization and resensitization. It binds to GPRK-phosphorylated receptors, sterically hindering their coupling to cognate G-proteins, thereby mediating homologous desensitization. As an endocytic adapter (CLASP), β-Arrestin 1 recruits GPCRs to the adapter protein 2 complex 2 (AP-2) in clathrin-coated pits (CCPs), facilitating receptor internalization. The extent of its involvement varies significantly depending on the receptor, agonist, and cell type. Class A receptors (e.g., ADRB2, OPRM1) dissociate from β-Arrestin 1 near the plasma membrane and undergo rapid recycling, while Class B receptors (e.g., AVPR2, AGTR1) internalize as complexes with β-Arrestin 1 and traffic to endosomal vesicles for prolonged periods. Additionally, β-Arrestin 1 acts as a signaling scaffold, activating pathways such as MAPK (e.g., ERK1/2), AKT1, and p38 MAPK, and participates in cytoskeletal rearrangement, chemotaxis, and transcriptional regulation. It also interacts with proteins like TRAF6 and ITCH to modulate Toll-like receptor, IL-1 receptor, and NOTCH signaling. by similar: Binds inositolhexakisphosphate (InsP6).
