Background

ZNF364 is an E3 ubiquitin-protein ligase that catalyzes 'Lys-48'- and/or 'Lys-63'-linked polyubiquitination of various substrates, playing roles in signaling pathways such as autophagy, innate immunity, cell proliferation, and cell death. It mediates endosomal trafficking and degradation of membrane receptors (e.g., EGFR, FLT3, MET, and CXCR4) through polyubiquitination. Collaborating with BST2, it contributes to antiviral immunity by facilitating HIV-1 virion internalization into intracellular vesicles for lysosomal degradation. Independently of BST2, it also exhibits antiviral activity by promoting ubiquitination, endo-lysosomal redistribution, and degradation of retroviral GAG proteins. During viral infection, ZNF364 differentially ubiquitinates MAVS ('Lys-48'-linked for proteasomal degradation) and STING1 ('Lys-63'-linked for oligomerization and TBK1 recruitment) to enhance innate antiviral responses. It positively regulates autophagosome-lysosome fusion by stabilizing STX17 without altering its ubiquitination levels, thereby promoting autophagosome maturation. Additionally, ZNF364 negatively regulates TLR-induced proinflammatory cytokine expression by catalyzing 'Lys-11'-linked ubiquitination of RAB1A and RAB13, inhibiting TLR trafficking from the ER to the Golgi (via RAB1A) and from the Golgi to the cell surface (via RAB13).
