Description

XRCC4 Antibody (YA5361) is a Mouse-derived and non-conjugated monoclonal antibody, targeting to XRCC4.

Background

XRCC4 is a core factor of DNA non-homologous end joining (NHEJ), essential for double-strand break repair and V(D)J recombination. It functions as a scaffold protein to regulate the recruitment of other proteins to DNA double-strand breaks (DSBs) and forms alternating helical filaments with NHEJ1/XLF, bridging and holding broken DNA together until repair is completed. The XRCC4-NHEJ1/XLF subcomplex binds DSB DNA fragments in a highly diffusive manner, robustly bridging them while keeping ends accessible for further processing. During the NHEJ ligation step of DSB repair, XRCC4 directly interacts with DNA ligase IV (LIG4) to form the LIG4-XRCC4 subcomplex, which reseals DNA breaks after gap filling. XRCC4 stabilizes LIG4, regulates its subcellular localization, and enhances its joining activity. It also promotes displacement of PNKP from processed break termini. by similar: XRCC4 acts as an activator of XKR4's phospholipid scramblase activity; upon caspase-3 (CASP3) cleavage, it translocates to the cytoplasm, interacts with XKR4, and induces phosphatidylserine exposure on apoptotic cell surfaces.
