Description

XPC Antibody (YA4158) is a mouse-derived and non-conjugated IgG1 antibody, targeting to XPC. It can be applicated for WB, IHC-P, ICC/IF, FC, ELISA assays, in the background of human.

Background

XPC functions in global genome nucleotide excision repair (GG-NER) as a damage-sensing and DNA-binding factor within the XPC complex. It exhibits low intrinsic DNA repair activity, which is enhanced by RAD23B and RAD23A. XPC preferentially binds DNA with short single-stranded segments rather than damaged oligonucleotides, a feature linked to its dynamic sensor role: it rapidly screens duplex DNA for non-hydrogen-bonded bases by forming transient nucleoprotein intermediates that mature into stable recognition complexes via single-stranded DNA-binding activity. The XPC complex is proposed to be the first factor binding DNA damage sites, partnering with XPA, RPA, and the TFIIH complex to form the pre-incision complex. It recognizes diverse helix-distorting lesions (e.g., single-stranded loops, mismatched bubbles, or overhangs), with binding orientation critical for productive NER. The complex detects unpaired bases on the undamaged strand, recruiting TFIIH for 5'→3' lesion scanning on the opposite strand. Cyclobutane pyrimidine dimers (CPDs) from UV damage evade XPC detection due to minimal structural distortion, requiring recognition by the UV-DDB complex for cooperative repair. In vitro, the XPC:RAD23B dimer alone initiates NER, preferentially binding cisplatin/UV-damaged dsDNA and diverse adducts while inducing DNA bending and stimulating TDG/SMUG1 glycosylase activity. Without DNA repair, XPC acts as a transcriptional coactivator: interacting with E2F1 at select promoters to recruit KAT2A and HAT complexes, specifically promoting acetylation of histone variant H2A.Z.1/H2A.Z (but not H2A.Z.2/H2A.V) to activate target genes.
