VPS35 Antibody (YA3314) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to VPS35.

Background

VPS35 is a component of the retromer cargo-selective complex (CSC), which serves as the core functional unit of retromer or its variants to prevent mis-sorting of specific transmembrane cargo proteins into the lysosomal degradation pathway. The CSC is recruited to endosomal membranes via RAB7A and SNX3. VPS35 primarily mediates cargo selectivity by interacting with the cytoplasmic domains of cargo proteins, though these interactions exhibit low affinity, and retromer SNX proteins may also contribute to cargo selection, challenging the classical CSC model. The SNX-BAR retromer facilitates retrograde transport of cargo proteins from endosomes to the trans-Golgi network (TGN) and participates in endosome-to-plasma membrane recycling. The SNX3-retromer mediates WLS transport from endosomes to the TGN via a distinct pathway from the SNX-BAR retromer. The SNX27-retromer is involved in endosome-to-plasma membrane trafficking and recycling of diverse cargo proteins. The CSC acts as a hub for recruiting other proteins, such as the WASH complex and TBC1D5. VPS35 is essential for retrograde transport of lysosomal enzyme receptors IGF2R and SLC11A2, regulates transcytosis of the polymeric immunoglobulin receptor (pIgR-pIgA), and is required for endosomal localization of WASHC2C and TBC1D5. It also mediates CSC association with the WASH complex via WASHC2. During microbial infection, the heterotrimeric retromer CSC facilitates human papillomavirus exit from early endosomes and delivery to the Golgi apparatus.

  • Immunohistochemical analysis of paraffin-embedded mouse colon tissue using VPS35 Antibody. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 8 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody at 1/100 dilution in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
