VPS24 is a probable core component of the endosomal sorting required for transport complex III (ESCRT-III), which is involved in the formation of multivesicular bodies (MVBs) and the sorting of endosomal cargo proteins into MVBs. MVBs contain intraluminal vesicles (ILVs) generated by invagination and scission from the endosomal membrane, primarily destined for lysosomal degradation of membrane proteins such as stimulated growth factor receptors, lysosomal enzymes, and lipids. The MVB pathway requires sequential action of ESCRT-O, -I, -II, and -III complexes. ESCRT-III proteins typically dissociate from the invaginating membrane before ILV release. The ESCRT machinery also participates in topologically equivalent membrane fission events, including cytokinesis and the budding of enveloped viruses (e.g., HIV-1 and other lentiviruses). ESCRT-III proteins, possibly with the AAA ATPase VPS4, mediate vesicle extrusion and/or membrane fission. VPS24 selectively binds phosphatidylinositol 3,5-bisphosphate PtdIns(3,5)P2 and PtdIns(3,4)P2 over other tested phosphoinositides. It functions in late cytokinesis and endosomal sorting/trafficking of the EGF receptor. by similar: Isoform 2, when expressed in yeast cells, prevents stress-induced cell death and reactive oxygen species accumulation.

  Western blot analysis was performed on extracts from 293T (lane 1, 15 μg), HepG2 (lane 2, 15 μg), U-87MG (lane 3, 15 μg), and 293 (lane 4, 15 μg) using VPS24 Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight.The primary antibody (1:2000 dilution) and the loading control antibody (beta-Actin, HY-P83730, 1:20000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti-Rabbit IgG-HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.
