VEGFR3 is a tyrosine-protein kinase that functions as a cell-surface receptor for VEGFC and VEGFD, playing a pivotal role in adult lymphangiogenesis and the development of the vascular network and cardiovascular system during embryonic development. It promotes the proliferation, survival, and migration of endothelial cells while regulating angiogenic sprouting. Activated FLT4 signaling enhances the production of VEGFC (and to a lesser extent VEGFA), establishing a positive feedback loop that amplifies FLT4 signaling. It modulates KDR signaling through heterodimer formation. The secreted isoform 3 may act as a decoy receptor for VEGFC and/or VEGFD, serving as a negative regulator of VEGFC-mediated lymphangiogenesis and angiogenesis. Binding of vascular growth factors to isoform 1 or isoform 2 triggers multiple signaling cascades; isoform 2 exhibits reduced signal transduction efficiency due to its truncated C-terminus and lack of key phosphorylation sites. VEGFR3 mediates activation of the MAPK1/ERK2, MAPK3/ERK1, MAPK8, JUN, and AKT1 signaling pathways. It phosphorylates SHC1 and facilitates the phosphorylation of PIK3R1, the regulatory subunit of phosphatidylinositol 3-kinase. Additionally, it promotes phosphorylation of MAPK8 at 'Thr-183' and 'Tyr-185' and AKT1 at 'Ser-473'.
