ULK1 Antibody (YA019) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to ULK1.

Background

ULK1 is a serine/threonine-protein kinase primarily involved in autophagy in response to starvation. It plays a crucial role in the formation of autophagophores, the precursors of autophagosomes, acting upstream of phosphatidylinositol 3-kinase PIK3C3. ULK1 is part of regulatory feedback loops in autophagy, functioning both as a downstream effector and negative regulator of mTORC1 via interaction with RPTOR. It is activated by AMPK-mediated phosphorylation and, in turn, regulates AMPK activity by phosphorylating its subunits PRKAA1, PRKAB2, and PRKAG1. ULK1 may also phosphorylate ATG13/KIAA0652 and RPTOR, though further evidence is needed. Additionally, ULK1 is essential for early neuronal differentiation and granule cell axon formation. It regulates autophagy by phosphorylating SESN2, SQSTM1, FLCN, and AMBRA1. Phosphorylation of AMBRA1 releases it from cytoskeletal docking sites to promote autophagosome nucleation. ULK1 also inhibits autophagy by phosphorylating ATG4B, reducing its proteolytic activation and delipidation activities.

  • Western blot analysis of extracts from MCF-7 (lane 2(20μg), MCF-7 (lane 3(40μg), using ULK1 Antibody. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in TBST for 2 hour at room temperature. The primary antibody and Loading control antibody (Beta Actin, HY-P80438, 1/3000) was used in 5% BSA in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (HY-P8004/HY-P8001, 1/10,000) was used for 1 hour at room temperature.
  • Immunohistochemical analysis of paraffin-embedded Mouse brain tissue using ULK1 Antibody. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 8.0) for 8 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (HY-P80366, 1/200) in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
