Background

UCP2 is an antiporter that exports dicarboxylate intermediates of the Krebs cycle from the mitochondrial inner membrane in exchange for phosphate and a proton, a process driven by mitochondrial motive force with broad effects on glycolysis, glutaminolysis, and glutathione-dependent redox balance. Continuous export of oxaloacetate and related four-carbon dicarboxylates from the mitochondrial matrix to the cytosol negatively regulates acetyl-CoA oxidation via the Krebs cycle, reducing the ATP/ADP ratio and reactive oxygen species (ROS) production. It may mediate inducible proton entry into the mitochondrial matrix, affecting ATP turnover as a protective mechanism against oxidative stress. Proton currents are likely linked to fatty acid flipping across the inner mitochondrial membrane, a process regulated by free fatty acids and purine nucleotides (By similarity). UCP2 regulates glucose utilization as an energy source and is required for glucose-induced DRP1-dependent mitochondrial fission and neuronal activation in the ventromedial hypothalamus (VMH). This mitochondrial adaptation mechanism modulates the VMH pool of glucose-excited neurons, influencing systemic glucose homeostasis (By similarity). During inflammation resolution, UCP2 controls macrophage ROS levels and metabolic reprogramming, attenuating IL33-induced ROS production to sustain Krebs cycle integrity for persistent itaconate synthesis and GATA3-dependent differentiation of inflammation-resolving macrophages (By similarity). UCP2 also unidirectionally transports anions, including L-malate, L-aspartate, phosphate, and chloride ions. Notably, UCP2 does not mediate adaptive thermogenesis (By similarity).
