Ubiquitin D Antibody (YA021) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to Ubiquitin D.

Ubiquitin D is a ubiquitin-like protein modifier that can be covalently attached to target proteins via adenylation of its C-terminal glycine (activated by UBA6), leading to their degradation by the 26S proteasome in a NUB1-dependent manner. It promotes the expression of the proteasome subunit beta type-9 (PSMB9/LMP2) and regulates TNF-alpha-induced and LPS-mediated activation of NF-kappa-B, a central mediator of innate immunity, by facilitating the proteasomal degradation of ubiquitinated I-kappa-B-alpha. In renal tubular epithelial cells (RTECs), Ubiquitin D is required for TNF-alpha-induced p65 nuclear translocation. It may also be involved in dendritic cell (DC) maturation, the process by which immature dendritic cells differentiate into fully competent antigen-presenting cells to initiate T-cell responses. In long-term in vitro culture and cancers, Ubiquitin D mediates mitotic non-disjunction and chromosome instability by abbreviating the mitotic phase and impairing the kinetochore localization of MAD2L1 during prometaphase. Additionally, it may participate in the formation of aggresomes when the proteasome is saturated or impaired. Ubiquitin D also mediates apoptosis in a caspase-dependent manner, particularly in renal epithelium and tubular cells during renal diseases such as polycystic kidney disease and HIV-associated nephropathy (HIVAN).

  Western blot analysis of extracts from Ramos using Ubiquitin D antibody. Proteins were transferred to a PVDF membrane and blocked with 5% nonfat powdered milk in PBST for 2 hour at room temperature. The primary antibody (1/1000) and loading control antibody (GAPDH, 1/3000) was diluted with 5% nonfat powdered milk in PBST at 4°C overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (1/8,000) was incubated for 45min at room temperature.
