UBA52 is a ubiquitin protein that can exist either covalently attached to other proteins or in a free (unanchored) form. When covalently bound, it conjugates to target proteins via an isopeptide bond as a monomer (monoubiquitin), a polymer linked through different Lys residues (polyubiquitin chains), or a linear polymer linked via the initiator Met (linear polyubiquitin chains). Polyubiquitin chains attached to target proteins have distinct functions depending on the linked Lys residue: Lys-6-linked may be involved in DNA repair; Lys-11-linked functions in ERAD (endoplasmic reticulum-associated degradation) and cell-cycle regulation; Lys-29-linked participates in proteotoxic stress response and cell cycle; Lys-33-linked is involved in kinase modification; Lys-48-linked mediates protein degradation via the proteasome; Lys-63-linked plays roles in endocytosis, DNA-damage responses, and signaling processes leading to NF-kappa-B activation. Linear polyubiquitin chains formed via the initiator Met contribute to cell signaling. Ubiquitin typically conjugates to Lys residues, but rare cases of conjugation to Cys or Ser have been observed. Free (unanchored) polyubiquitin also has distinct roles, such as activating protein kinases and signaling. Additionally, UBA52 is a component of the 60S ribosomal subunit, where ribosomal protein L40 is essential for translating a subset of cellular transcripts, particularly cap-dependent translation of vesicular stomatitis virus mRNAs.
