TNF α Antibody (YA6105) is a rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to TNF-α. It can be applicated for WB, IHC-P, ICC/IF, IP, ELISA assays, in the background of human, mouse, rat.

TNF α is a cytokine that binds to TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. Primarily secreted by macrophages, it induces cell death in certain tumor cell lines and acts as a potent pyrogen, triggering fever directly or via interleukin-1 secretion stimulation, while also implicated in cachexia induction. Under specific conditions, it stimulates cell proliferation and differentiation. In rheumatoid arthritis, it impairs regulatory T-cell (Treg) function by FOXP3 dephosphorylation, upregulating protein phosphatase 1 (PP1) to inactivate FOXP3 at the 'Ser-418' residue, rendering Tregs dysfunctional. TNF α is a key mediator of cell death in BCG-stimulated neutrophils combined with DIABLO/SMAC mimetic therapy for RT4v6 bladder cancer. It induces insulin resistance in adipocytes by inhibiting insulin-induced IRS1 tyrosine phosphorylation and glucose uptake (by similarity: partially via TNF-triggered GKAP42 degradation). Synergistically with IL1B and IL6, it promotes VEGF production for angiogenesis and osteoclastogenesis-mediated bone resorption (by similarity). Its intracellular domain (ICD) form stimulates IL12 production in dendritic cells.
