Background

TIMELESS is a protein that plays a crucial role in controlling DNA replication, maintaining replication fork stability, preserving genome stability during normal DNA replication, DNA repair, and regulating the circadian clock. It stabilizes replication forks by forming a complex with TIPIN, regulating DNA replication under both normal and stress conditions, and influencing CHEK1 phosphorylation and the intra-S phase checkpoint. TIMELESS inhibits the CMG complex's ATPase activity and activates DNA polymerases, coupling DNA unwinding and synthesis. It promotes TIPIN nuclear localization and aids in processive DNA replication past G-quadruplex (G4) structures, possibly with DDX1. The protein is involved in cell survival after DNA damage or replication stress by facilitating DNA repair and, in response to double-strand breaks (DSBs), accumulates at damage sites to promote homologous recombination repair via PARP1 interaction. TIMELESS may also be essential for the ATR-CHEK1 pathway in hydroxyurea- or UV-induced replication checkpoints. It contributes to circadian clock regulation by determining period length and advancing the clock phase in response to DNA damage. Additionally, it negatively regulates CLOCK|NPAS2-ARTNL/BMAL1|ARTNL2/BMAL2-induced PER1 transactivation, possibly by translocating PER1 into the nucleus, and may destabilize the PER2-CRY2 complex. By similarity, TIMELESS might also play a role in epithelial cell morphogenesis and branching tubule formation.
