Description

TIM3 Antibody (YA4380) is a mouse-derived and non-conjugated IgG1 antibody, targeting to TIM3. It can be applicated for ICC/IF, FC, ELISA assays, in the background of human.

Background

TIM3 is a cell surface receptor implicated in modulating innate and adaptive immune responses, primarily recognized for its inhibitory role, though its activity may vary depending on cellular context or specific ligands like LGALS9. It regulates macrophage activation, suppresses Th1-mediated auto- and alloimmune responses, and promotes tolerance. In CD8+ T cells, TIM3 attenuates TCR signaling by blocking NF-kappaB and NFAT promoter activities, reducing IL-2 secretion, potentially via LCK association or LGALS9-dependent recruitment of PTPRC to the immunological synapse. Conversely, similar studies suggest TIM3 may activate TCR signaling in T-cells through ZAP70, LCP2, LCK, and FYN. Expressed on Tregs, it inhibits Th17 responses. As a receptor for LGALS9, TIM3 binding suppresses T-cell responses and induces apoptosis of antigen-specific cells via HAVCR2 phosphorylation and disrupted BAG6 interaction. It may also contribute to innate immunity against intracellular pathogens, such as by enhancing antibacterial activity (e.g., IL-1β secretion) in Mycobacterium tuberculosis-infected macrophages (By similarity), though its role as a LGALS9 receptor is debated. TIM3 also binds phosphatidylserine (PtSer), mediating phagocytosis of apoptotic cells by dendritic cells (DCs). In DCs, it synergizes with Toll-like receptors to promote TNF-alpha secretion, while in tumor-infiltrating DCs, it suppresses nucleic acid sensing by interacting with HMGB1 (By similarity). On NK cells, TIM3 acts as a coreceptor to boost LGALS9-induced IFN-gamma production but may suppress cytotoxicity and negatively regulate NK function during endotoxic shock (By similarity).
