Background

Tight Junction Protein ZO 3 (TJP3) is a scaffolding protein closely related to TJP1 and TJP2, linking tight junction (TJ) transmembrane proteins such as claudins, junctional adhesion molecules, and occludin to the actin cytoskeleton. The tight junction functions to limit paracellular movement of substances and serves as a boundary between the distinct apical and basolateral plasma membrane domains of epithelial and endothelial cells. TJP3 binds and recruits PATJ to tight junctions, stabilizing apical and lateral components. It also promotes cell-cycle progression by sequestering cyclin D1 (CCND1) at tight junctions during mitosis, preventing its degradation and enabling S-phase transition. Similar to TJP1 and TJP2, TJP3 contributes to the junctional retention and stability of the transcription factor DBPA but is not involved in its nuclear shuttling. Unlike TJP2, TJP3 is dispensable for viability, embryonic development, epithelial differentiation, and TJ establishment, at least under laboratory conditions.

  • Western blot analysis of extracts from Caco-2 (lane 2(20μg)) using Zonula occludens protein 3 Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (beta Actin, HY-P83730, 1/5000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Rabbit/Mouse IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.
