Thrombospondin 1 is an adhesive glycoprotein that mediates cell-to-cell and cell-to-matrix interactions, with multifunctional roles in inflammation, angiogenesis, wound healing, reactive oxygen species (ROS) signaling, nitrous oxide (NO) signaling, apoptosis, senescence, cellular self-renewal, stemness, and cardiovascular and metabolic homeostasis. It negatively modulates dendritic cell activation and cytokine release via CD47, contributing to immune homeostasis. Additionally, Thrombospondin 1 regulates NO signaling through CD47, acting as a pressor agent to support blood pressure (By similarity). It promotes endothelial cell senescence by increasing NADPH oxidase NOX1 activity and ROS generation via CD47. The protein also inhibits stem cell self-renewal (By similarity) and negatively regulates wound healing (By similarity) through CD47 signaling. By binding to CD36, it induces podocyte apoptosis in response to elevated free fatty acids (By similarity) and suppresses angiogenesis via CD36 or CD47 depending on context. Thrombospondin 1 stimulates ROS production in non-phagocytic cells through interaction with SIRPA and may exacerbate adipose inflammatory activity in diet-induced obesity (By similarity). It participates in ER stress response via ATF6 interaction (By similarity) and is implicated in age-related metabolic dysregulation.
