TFIIH p89 Antibody (YA5168) is a mouse-derived and non-conjugated monoclonal antibody, targeting to TFIIH p89. It can be applicated for WB assays, in the background of human, mouse, rat, bovine, dog.

Background

TFIIH p89 is an ATP-dependent 3'-5' DNA helicase/translocase that binds dsDNA rather than ssDNA, unwinding it via translocase activity rather than classical helicase function. As a core component of the general transcription and DNA repair factor IIH (TFIIH) complex, it participates in RNA polymerase II-mediated transcription when associated with CDK-activating kinase (CAK). Its ATPase activity (but not helicase activity) is required for DNA opening, potentially by wrapping damaged DNA to wedge it open, creating localized melting for XPD/ERCC2 helicase anchoring. In transcription, TFIIH is essential for initiation, inducing DNA twisting to open templates, with its ATP-dependent helicase activity critical for promoter opening/escape. It also functions in transcription-coupled nucleotide excision repair (NER) by unwinding damaged DNA to enable excision and replacement. The NER-TFIIH complex exhibits structural differences from the transcription complex, with XPA stabilizing large movements of XPD/ERCC2 and XPB/ERCC3 while anchoring the latter at the 5' end of DNA bubbles.
