Background

TBK1 is a serine/threonine kinase that plays a crucial role in regulating inflammatory responses to pathogens. Upon activation of toll-like receptors by viral or bacterial components, TBK1 associates with TRAF3 and TANK, phosphorylating interferon regulatory factors (IRFs) IRF3 and IRF7 as well as DDX3X, leading to IRF dimerization, nuclear translocation, and transcriptional activation of pro-inflammatory and antiviral genes such as IFNA and IFNB. TBK1 licenses IRF3 for phosphorylation by first modifying adapter proteins MAVS, STING1, and TICAM1 at their pLxIS motifs. It also regulates NF-kappa-B signaling by phosphorylating NFKBIA, IKBKB, or RELA. In autophagy, TBK1 phosphorylates OPTN/Optineurin to enhance LC3 binding and antibacterial autophagy, while also targeting SMCR8 (part of the C9orf72-SMCR8 complex) and ATG8 proteins (e.g., MAP1LC3C and GABARAPL2) to regulate autophagosome maturation. In energy balance, TBK1 may sustain obesity-related chronic inflammation, impairing insulin sensitivity (By similarity). Additionally, TBK1 attenuates retroviral budding, modulates mTORC1 and mTORC2 activity, influences TNF-induced RIPK1-mediated cell death, and participates in T follicular regulatory cell differentiation.
