TAK1 is a serine/threonine kinase that serves as a critical component of the MAP kinase signal transduction pathway. It plays a vital role in cellular response cascades triggered by environmental changes, mediating signal transduction for TRAF6, various cytokines (including IL-1, TGFB, and related factors like BMP2 and BMP4), toll-like receptors (TLR), tumor necrosis factor receptor CD40, and the B-cell receptor (BCR). Upon activation, TAK1 acts as an upstream activator of the MKK/JNK and p38 MAPK signaling cascades by phosphorylating and activating MAP kinase kinases such as MAP2K1/MEK1, MAP2K3/MKK3, MAP2K6/MKK6, and MAP2K7/MKK7. These pathways regulate the transcription factor AP-1. Independently of MAP2Ks and p38 MAPKs, TAK1 is a key activator of NF-kappa-B by promoting the activation of the IKK core complex. In osmotic stress signaling, TAK1 primarily activates MAPK8/JNK1 without affecting NF-kappa-B. It also enhances TRIM5's capsid-specific restriction activity. By similarity, TAK1 phosphorylates RIPK1 at Ser-321, positively regulating RIPK1-RIPK3 interaction to promote necroptosis while negatively regulating RIPK1 kinase activity and its interaction with FADD to mediate apoptosis. Additionally, TAK1 phosphorylates STING1 in response to cGAMP activation, facilitating the association between STEEP1 and STING1 and STING1 translocation to COPII vesicles.
