Background

SUN1 is a component of the LINC (LInker of Nucleoskeleton and Cytoskeleton) complex, which mediates the connection between the nuclear lamina and the cytoskeleton. The nucleocytoplasmic interactions established by the LINC complex play a key role in transmitting mechanical forces across the nuclear envelope and in nuclear movement and positioning (by similar). SUN1 is required for interkinetic nuclear migration (INM) and is essential for nucleokinesis and centrosome-nucleus coupling during radial neuronal migration in the cerebral cortex and glial migration (by similar). It is involved in telomere attachment to the nuclear envelope during meiotic prophase, functioning as part of a SUN1/2:KASH5 LINC complex where SUN1 and SUN2 appear to act at least partially redundantly (by similar). SUN1 is necessary for gametogenesis and contributes to the selective expression of coding and non-coding RNAs required for gametogenesis (by similar). It helps define the distribution of nuclear pore complexes (NPCs) (by similar) and is required for efficient localization of SYNE4 at the nuclear envelope (by similar). SUN1 may also participate in nuclear remodeling during sperm head formation in spermatogenesis (by similar). Additionally, it may play a role in DNA repair by suppressing non-homologous end joining to facilitate the repair of DNA cross-links.

  • Western blot analysis was performed on extracts from HeLa (lane 1, 20 μg), A431 (lane 2, 20 μg), and Ramos (lane 3, 20 μg) using SUN1 Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non - fat milk in TBST at 4°C overnight.The primary antibody (1:1000 dilution) and the loading control antibody (GAPDH, HY-P2804, 1:5000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti - Rabbit IgG - HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.
