Stromal Interaction Molecule 1 (STIM1) acts as a Ca²⁺ sensor that gates two major inward rectifying Ca²⁺ channels at the plasma membrane: Ca²⁺ release-activated Ca²⁺ (CRAC) channels and arachidonate-regulated Ca²⁺-selective (ARC) channels. It plays a key role in mediating store-operated Ca²⁺ entry (SOCE), a Ca²⁺ influx triggered by depletion of intracellular Ca²⁺ stores. Upon Ca²⁺ depletion, STIM1 translocates from the endoplasmic reticulum to the plasma membrane, where it activates CRAC channel pore-forming subunits ORA1, ORA2, and ORAI3 to generate sustained and oscillatory Ca²⁺ entry.
