SREBP-1 Antibody (YA5498) is a mouse-derived and non-conjugated monoclonal antibody, targeting to SREBP-1. It can be applicated for WB, ICC/IF, ELISA assays, in the background of human, mouse, rat.

Background

SREBP-1 is the precursor of the transcription factor form (Processed sterol regulatory element-binding protein 1), embedded in the endoplasmic reticulum membrane. Low sterol concentrations promote its processing, releasing the transcription factor form that translocates into the nucleus and activates transcription of genes involved in cholesterol biosynthesis and lipid homeostasis (By similarity). As a key transcription factor, it regulates the expression of genes related to cholesterol biosynthesis and lipid homeostasis, binding to the sterol regulatory element 1 (SRE-1, 5'-ATCACCCCAC-3') and exhibiting dual sequence specificity for both an E-box motif (5'-ATCACGTGA-3') and SRE-1 (5'-ATCACCCCAC-3'). It regulates promoters of genes involved in cholesterol biosynthesis and the LDL receptor (LDLR) pathway. The isoform expressed in select tissues (SREBP-1A) has higher transcriptional activity compared to SREBP-1C (By similarity), stimulating both lipogenic and cholesterogenic gene expression and playing a role in the nutritional regulation of fatty acids and triglycerides in lipogenic organs such as the liver (By similarity). It is also required for innate immune responses in macrophages by regulating lipid metabolism (By similarity). The predominant isoform in most tissues (SREBP-1C) has weaker transcriptional activity (By similarity), primarily controlling lipogenic gene expression and strongly activating global lipid synthesis in rapidly growing cells (By similarity). Certain isoforms are constitutively active in transactivating lipogenic gene promoters without requiring Golgi proteolytic processing.
