SRC1 is a nuclear receptor coactivator that directly binds nuclear receptors and stimulates transcriptional activity in a hormone-dependent manner. It participates in the coactivation of various nuclear receptors, including those for steroids (e.g., PGR, GR, and ER), retinoids (e.g., RXRs), thyroid hormone (e.g., TRs), and prostanoids (e.g., PPARs). It is also involved in coactivation mediated by STAT3, STAT5A, STAT5B, and STAT6 transcription factors. SRC1 exhibits histone acetyltransferase activity toward H3 and H4, though the functional relevance of this activity remains unclear. It plays a central role in assembling multisubunit coactivator complexes that act via chromatin remodeling and may participate in both chromatin remodeling and the recruitment of general transcription factors. Together with NCOA2, it regulates energy balance between white and brown adipose tissues and is essential for mediating steroid hormone responses. Isoform 2 demonstrates higher thyroid hormone-dependent transactivation activity compared to isoforms 1 and 3.
