Description	SQSTM1/p62 Antibody (YA669) is a Mouse-derived and non-conjugated IgG1 monoclonal antibody, targeting to SQSTM1/p62.
Background	SQSTM1 is a molecular adapter required for selective macroautophagy (aggrephagy) by acting as a bridge between polyubiquitinated proteins and autophagosomes. It promotes the recruitment of ubiquitinated cargo proteins to autophagosomes via multiple domains that bridge proteins and organelles in different steps. SQSTM1 first mediates the assembly of ubiquitinated proteins by undergoing liquid-liquid phase separation via its UBA domain, forming insoluble cytoplasmic inclusions called p62 bodies. It then interacts with ATG8 family proteins on autophagosomes via its LIR motif, leading to p62 body recruitment and autophagic clearance of ubiquitinated proteins. SQSTM1 is itself degraded along with its ubiquitinated cargos. Additionally, SQSTM1 recruits ubiquitinated proteins to PML bodies in the nucleus and participates in peroxisome autophagy (pexophagy) by bridging ubiquitinated PEX5 and autophagosomes in response to reactive oxygen species (ROS). It also activates the NFE2L2/NRF2 pathway by sequestering KEAP1, promoting nuclear accumulation of NFE2L2/NRF2 and cytoprotective gene expression. SQSTM1 facilitates relocalization of 'Lys-63'-linked ubiquitinated STING1 to autophagosomes and organizes endosomes by retaining vesicles in the perinuclear cloud via RNF26-mediated ubiquitination. It may regulate NFKB1 activation by TNF-alpha, nerve growth factor (NGF), and interleukin-1, and mediates the interaction between TRAF6 and CYLD (by similar).
