Description

SPHK1 Antibody (YA063) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to SPHK1.

Background

SPHK1 catalyzes the phosphorylation of sphingosine to form sphingosine 1-phosphate (SPP), a lipid mediator with intra- and extracellular functions. It primarily acts on D-erythro-sphingosine and, to a lesser extent, sphinganine, but not other lipids such as D,L-threo-dihydrosphingosine, N,N-dimethylsphingosine, diacylglycerol, ceramide, or phosphatidylinositol. Unlike the proapoptotic SPHK2, SPHK1 reduces intracellular ceramide levels, promotes cell growth, and inhibits apoptosis. It regulates inflammatory responses and neuroinflammation by stimulating TRAF2 E3 ubiquitin ligase activity via SPP, activating NF-kappa-B in TNF signaling to induce IL17 secretion. In response to TNF, SPHK1 negatively regulates RANTES induction through the p38 MAPK pathway. It also participates in sphingosine-induced endocytic membrane trafficking, localizing to dilated endosomes and functioning in later stages of endosomal maturation and membrane fusion (independent of its kinase activity). In Purkinje cells, SPHK1 may regulate autophagosome-lysosome fusion upon VEGFA stimulation. by similar: SPHK1 exhibits acetyl-CoA-dependent serine acetyltransferase activity on PTGS2/COX2, enhanced in the presence of sphingosine. During neuroinflammation, it promotes neuronal secretion of specialized pro-resolving mediators (SPMs), particularly 15-R-lipoxin A4, through PTGS2 acetylation, increasing microglial phagocytosis.

  • Western blot analysis of extracts from HEK293(lane 2(20μg) , HEK293(lane 3(40μg) and MCF-7(lane 4(20ug) ,MCF-7(lane 5(40ug) using SPHK1(HY-P80337Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.
  • Flow cytometric analysis of 1X10^6 Raji cells labeling SPHK1 Antibody (HY-P80337, red). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/50 dilution for an hour at 4℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L (HY-P8002) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Rabbit IgG Isotype Control (HY-P80879, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
