Description

SOX9 Antibody (YA4046) is a mouse-derived and non-conjugated IgG1 antibody, targeting to SOX9. It can be applicated for WB, ICC/IF, ELISA assays, in the background of human.

Background

SOX9 is a transcription factor that plays a key role in chondrocyte differentiation and skeletal development. It specifically binds the 5'-ACAAAG-3' DNA motif in enhancers and super-enhancers, promoting the expression of genes critical for chondrogenesis, including cartilage matrix-coding genes COL2A1, COL4A2, COL9A1, COL11A2, and ACAN, as well as SOX5 and SOX6. By similarity, it also binds to some promoter regions and acts centrally in successive steps of chondrocyte differentiation. It is absolutely required for precartilaginous condensation, the first step in chondrogenesis where skeletal progenitors differentiate into prechondrocytes. Together with SOX5 and SOX6, it drives overt chondrogenesis as prechondrocytes differentiate into early-stage chondrocytes. Later, it directs hypertrophic maturation and blocks osteoblast differentiation in growth plate chondrocytes by maintaining columnar proliferation, delaying prehypertrophy, and suppressing osteoblastic differentiation via reduced β-catenin (CTNNB1) signaling and RUNX2 expression. By similarity, it also supports chondrocyte hypertrophy both indirectly (by preserving chondrocyte lineage fate) and directly (by transactivating COL10A1 with MEF2C in upper hypertrophic cells). Low lipid levels trigger chondrogenic commitment via FOXO (FOXO1/FOXO3)-mediated SOX9 upregulation, which suppresses fatty acid oxidation. By similarity, it aids (but is not essential for) removing repressive epigenetic marks and depositing active chromatin signatures at chondrocyte-specific genes. It cooperates with Hedgehog-dependent GLI (GLI1/GLI3) factors. Beyond cartilage, SOX9 regulates proliferation and differentiation in epithelial stem/progenitor cells, balancing these processes during lung branching morphogenesis and extracellular matrix regulation. By similarity, it also controls epithelial branching in kidney development.
