SMC1 is a central component of the cohesin complex, functioning in chromosome cohesion during the cell cycle and DNA repair. The cohesin complex maintains sister chromatid cohesion after DNA replication, potentially forming a large proteinaceous ring structure to entrap sister chromatids. During anaphase, the complex is cleaved and dissociates from chromatin to enable chromatid separation. It may also contribute to mitotic spindle pole assembly. For DNA repair, SMC1 operates through two distinct pathways: 1) interaction with BRCA1 coupled with ATM-mediated phosphorylation, and 2) ATR-dependent phosphorylation. It serves as a downstream effector in both the ATM/NBS1 and ATR/MSH2 branches of the S-phase checkpoint.
