SMARCA1 Antibody (YA3932) is a mouse-derived and non-conjugated IgG1 antibody, targeting to SMARCA1. It can be applicated for WB, IHC-P, FC, ELISA assays, in the background of human, mouse.

Background

SMARCA1 is an ATPase with intrinsic ATP-dependent chromatin-remodeling activity (PubMed:14609955, PubMed:15310751, PubMed:15640247, PubMed:28801535). Its ATPase activity is substrate-dependent, increasing when nucleosomes are the substrate, though it remains catalytically active with DNA alone (PubMed:14609955, PubMed:15310751, PubMed:15640247). As the catalytic subunit of ISWI chromatin-remodeling complexes, it forms ordered nucleosome arrays on chromatin, facilitating DNA accessibility during processes like replication, transcription, and repair (PubMed:14609955, PubMed:15310751, PubMed:15640247, PubMed:28801535). Within these complexes, SMARCA1 slides edge- and center-positioned histone octamers away from their original DNA locations (PubMed:28801535). Its catalytic activity and sliding propensity are regulated by ISWI complex components (PubMed:28801535). BAZ1A-, BAZ1B-, BAZ2A-, and BAZ2B-containing ISWI complexes regulate nucleosome spacing and can slide mononucleosomes to DNA template centers (PubMed:28801535), whereas CECR2- and RSF1-containing complexes lack this ability (PubMed:28801535). In the NURF-1 and CERF-1 ISWI complexes, nucleosomes are the preferred ATPase substrate (PubMed:14609955, PubMed:15640247). Within NURF-1, SMARCA1 binds En1 and En2 promoters to upregulate their expression and support brain development (PubMed:14609955). It may promote neurite outgrowth (PubMed:14609955) and luteal cell development (PubMed:16740656). During DNA replication, it facilitates nucleosome assembly, ensuring fork progression and genomic stability by preventing replication stress and nascent DNA gaps (PubMed:39413208). SMARCA1 is catalytically inactive with DNA or nucleosome substrates and lacks remodeling activity in these contexts (PubMed:15310751, PubMed:28801535), acting as a negative regulator by forming inactive complexes (PubMed:15310751).
