PTPN11 is a protein tyrosine phosphatase that acts downstream of various receptor and cytoplasmic protein tyrosine kinases, participating in signal transduction from the cell surface to the nucleus. It positively regulates the MAPK signaling pathway and dephosphorylates GAB1, ARHGAP35, and EGFR. Additionally, PTPN11 dephosphorylates ROCK2 at Tyr-722 to enhance its RhoA binding activity and dephosphorylates CDC73. By similarity, it dephosphorylates tyrosine residues on SOX9, inactivating SOX9 and promoting ossification. Experimental evidence also shows its ability to dephosphorylate tyrosine-phosphorylated NEDD9/CAS-L.
