SESN2 Antibody (YA5993) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to SESN2.

SESN2 functions as an intracellular leucine sensor that negatively regulates the mTORC1 signaling pathway via the GATOR complex. In the absence of leucine, it binds the GATOR2 subcomplex to inhibit mTORC1 signaling; leucine binding disrupts this interaction, activating the TORC1 pathway. This stress-inducible metabolic regulator also protects against oxidative and genotoxic stress. It may negatively regulate protein translation during ER stress through mTORC1 and positively regulate NFE2L2-mediated oxidative stress response gene transcription by promoting SQSTM1-dependent autophagic degradation of KEAP1. SESN2 may also mediate TP53's inhibition of TORC1 signaling under genotoxic stress and prevent ROS accumulation via its N-terminal alkylhydroperoxide reductase activity. While initially reported to enhance oxidative stress resistance by reducing PRDX1, this finding was later unconfirmed.
