Background

SEH1L is a component of the Nup107-160 subcomplex of the nuclear pore complex (NPC), which is essential for the assembly of a functional NPC and plays a role in kinetochore microtubule attachment, mitotic progression, and chromosome segregation. Additionally, SEH1L contributes to the recruitment of the Nup107-160 subcomplex to the kinetochore. As part of the GATOR2 complex, SEH1L functions as an activator of the amino acid-sensing branch of the mTORC1 signaling pathway by inhibiting the GATOR1 subcomplex. In the presence of abundant amino acids, GATOR2 mediates ubiquitination of NPRL2, a core component of GATOR1, leading to its inactivation. Conversely, under amino acid deprivation, GATOR2 is inhibited, activating GATOR1. Within the GATOR2 complex, SEC13 and SEH1L are required for complex stabilization.
