Description

SARS-COV-2 Spike Antibody (YA5389) is a Mouse-derived and non-conjugated monoclonal antibody, targeting to SARS-COV-2 Spike.

Background

The SARS-COV-2 Spike protein attaches the virion to the cell membrane by interacting with host receptors, primarily ACE2, to initiate infection. When S2/S2' is cleaved, receptor binding triggers direct fusion at the cell membrane; if uncleaved, it leads to virus internalization via host TFRC and GRM2-mediated endocytosis, resulting in virion-endosomal membrane fusion. Alternatively, it may use NRP1/NRP2 and integrins as entry receptors, with NRP1/NRP2 potentially explaining viral tropism in human olfactory epithelial cells. The stalk domain of Spike contains three hinges, providing head orientational flexibility. As a precursor of the fusion protein, it mediates membrane fusion as a class I viral fusion protein, containing two unmasked fusion peptides after cleavage. S2/S2' cleavage occurs either at the cell membrane by host TMPRSS2 or during endocytosis by host CSTL, triggering irreversible conformational changes that fuse viral and cellular membranes, releasing viral RNA into the cytoplasm. The protein exhibits at least three states: pre-fusion native, pre-hairpin intermediate, and post-fusion hairpin, with coiled-coil regions forming a trimer-of-hairpins structure during fusion to position the fusion peptide near the ectodomain's C-terminus, facilitating membrane apposition and fusion.
