Background

S100A9 is a calcium- and zinc-binding protein that plays a prominent role in regulating inflammatory processes and immune responses. It induces neutrophil chemotaxis, adhesion, and enhances bactericidal activity by promoting phagocytosis through activation of SYK, PI3K/AKT, and ERK1/2, as well as triggering degranulation via a MAPK-dependent mechanism (By similarity). Predominantly found as calprotectin (S100A8/A9), it exhibits diverse intracellular and extracellular functions (By similarity). Intracellularly, it facilitates leukocyte arachidonic acid trafficking and metabolism, modulates the tubulin-dependent cytoskeleton during phagocyte migration, and activates neutrophilic NADPH oxidase. It also collaborates with CD69 in regulatory T-cell differentiation (By similarity). Extracellularly, it displays pro-inflammatory, antimicrobial, oxidant-scavenging, and apoptosis-inducing activities. Its pro-inflammatory effects include leukocyte recruitment, cytokine/chemokine production, and regulation of leukocyte adhesion and migration (By similarity). As an alarmin or DAMP molecule, it stimulates innate immunity by binding to pattern recognition receptors like TLR4 and AGER, activating MAPK and NF-kappa-B pathways to amplify inflammation. Its antimicrobial action likely involves Zn2+ chelation (By similarity). S100A9 can induce autophagy- and apoptosis-mediated cell death through mitochondrial-lysosomal crosstalk involving ROS and BNIP3 (By similarity). It regulates neutrophil survival via ITGAM/ITGB-TLR4-MEK-ERK signaling (By similarity) and protects tissues by scavenging oxidants (By similarity). The iNOS-S100A8/A9 transnitrosylase complex selectively S-nitrosylates targets like GAPDH and VIM by recognizing an [IL]-x-C-x-x-[DE] motif (By similarity).
