S100A9 is a calcium- and zinc-binding protein that plays a prominent role in regulating inflammatory processes and immune responses. It induces neutrophil chemotaxis, adhesion, and enhances bactericidal activity by promoting phagocytosis through activation of SYK, PI3K/AKT, and ERK1/2, as well as triggering degranulation via a MAPK-dependent mechanism (By similarity). Predominantly found as calprotectin (S100A8/A9), it exhibits diverse intracellular and extracellular functions (By similarity). Intracellularly, it facilitates leukocyte arachidonic acid trafficking and metabolism, modulates the tubulin-dependent cytoskeleton during phagocyte migration, and activates neutrophilic NADPH oxidase. It also collaborates with CD69 in regulatory T-cell differentiation (By similarity). Extracellularly, it displays pro-inflammatory, antimicrobial, oxidant-scavenging, and apoptosis-inducing activities.
