Description

RUNX1 Antibody (YA5052) is a mouse-derived and non-conjugated monoclonal antibody, targeting to RUNX1. It can be applicated for WB, ICC/IF, ELISA assays, in the background of human.

Background

RUNX1 forms the heterodimeric complex core-binding factor (CBF) with CBFB. RUNX family members modulate target gene transcription by recognizing the core consensus binding sequence 5'-TGTGGT-3' (or rarely 5'-TGCGGT-3') in regulatory regions via their runt domain, while CBFB is a non-DNA-binding regulatory subunit that allosterically enhances RUNX's DNA-binding specificity. The heterodimer binds core sites of various enhancers and promoters, including murine leukemia virus, polyomavirus enhancer, T-cell receptor enhancers, LCK, IL3, and GM-CSF promoters. Essential for normal hematopoiesis, RUNX1 synergizes with ELF4 to transactivate the IL-3 promoter and with ELF2 for the BLK promoter. It inhibits KAT6B-dependent transcriptional activation (by similarity) and regulates lineage commitment of immature T-cell precursors. CBF complexes repress ZBTB7B during cytotoxic (CD8+) T-cell development by binding RUNX sites in the ZBTB7B locus, acting as a transcriptional silencer to enable cytotoxic T-cell differentiation. RUNX1 also controls regulatory T-cell (Treg) anergy and suppressive function via FOXP3 association, activating IL2 and IFNG while downregulating TNFRSF18, IL2RA, and CTLA4 in conventional T-cells (by similarity). In T-helper 17 cells, it positively regulates RORC expression. Isoform AML-1G exhibits higher binding affinity for target genes (e.g., TCR-beta-E2 and RAG-1 sites) but is less effective in neutrophil differentiation. Isoform AML-1L interferes with RUNX1 transactivation activity.
