RPA70 Antibody (YA5280) is a Mouse-derived and non-conjugated monoclonal antibody, targeting to RPA70.

RPA70 is a component of the heterotrimeric replication protein A complex (RPA/RP-A), which binds and stabilizes single-stranded DNA intermediates formed during DNA replication or under DNA stress, preventing their reannealing while recruiting and activating various proteins and complexes involved in DNA metabolism. It plays a crucial role in both DNA replication and the cellular response to DNA damage. In the DNA damage response, the RPA complex regulates DNA repair and checkpoint activation by recruiting ATRIP to activate ATR kinase, a master regulator of this process. It facilitates the recruitment of double-strand break repair factors RAD51 and RAD52 to chromatin and recruits proteins like XPA and XPG for nucleotide excision repair (NER). Additionally, RPA70 likely participates in base excision repair (BER) through interaction with UNG and recruits SMARCAL1/HARP to promote replication fork restart. It also contributes to telomere maintenance. As part of the alternative RPA complex (aRPA), it binds single-stranded DNA and may function in DNA repair. Unlike the canonical RPA complex containing RPA2, aRPA may not support chromosomal DNA replication or S-phase progression but can assist DNA synthesis by polymerase delta in the presence of PCNA and RFC, as well as the dual incision/excision reaction in NER and RAD51-dependent strand exchange. RPA also stimulates the 5'-3' helicase activity of BRIP1/FANCJ.
