Ran Antibody (YA4129) is a mouse-derived and non-conjugated IgG1 antibody, targeting to RAN. It can be applicated for WB, IHC-P, ICC/IF, FC, ELISA assays, in the background of human, mouse, monkey, rat.

RAN is a GTPase involved in nucleocytoplasmic transport, facilitating both the import and export of proteins and RNAs between the nucleus and cytoplasm. It switches between a cytoplasmic GDP-bound state and a nuclear GTP-bound state via nucleotide exchange and GTP hydrolysis. Nuclear import receptors like importin beta bind their cargo only in the absence of GTP-bound RAN and release it upon interaction with GTP-bound RAN, whereas export receptors operate inversely. This mechanism ensures compartment-specific cargo loading/unloading and directional transport. Interaction with RANBP1 induces a conformational change in the XPO1-RAN complex, triggering the release of cargo nuclear export signals. GTP-bound RAN promotes microtubule assembly at mitotic chromosomes and is essential for proper spindle formation and chromosome segregation during mitosis. It also regulates mitotic progression. The RAN-BIRC5/survivin complex aids spindle assembly by scaffolding the delivery of the effector TPX2 to microtubules. Additionally, RAN negatively regulates VRK1/VRK2 kinase activity and enhances AR-mediated transactivation (which diminishes with increasing poly-Gln length in AR).
