RAD23B is a multifunctional protein primarily involved in modulating proteasomal degradation and nucleotide excision repair (NER). As a polyubiquitin chain receptor, it binds to polyubiquitin chains and simultaneously interacts with the 26S proteasome and ubiquitinated substrates, delivering ubiquitinated proteins for proteasomal degradation. In endoplasmic reticulum-associated degradation (ERAD), it collaborates with PNGase to target deglycosylated proteins to the proteasome. Within global genome NER (GG-NER), it acts as a component of the XPC complex, stabilizing XPC in cooperation with CETN2 and potentially protecting XPC from proteasomal degradation. The XPC complex serves as the initial damage recognition factor, forming a pre-incision complex with XPA, RPA, and the TFIIH complex to detect DNA helix distortions (e.g., single-stranded loops, mismatched bubbles). The XPC:RAD23B dimer alone can initiate NER in vitro, preferentially binding cisplatin- and UV-damaged DNA, as well as diverse DNA adducts. It also stimulates the activity of DNA glycosylases TDG and SMUG1.
