PTH/PTHrP-R is a G-protein-coupled receptor that binds parathyroid hormone (PTH) and parathyroid hormone-related peptide (PTHLH). Ligand binding induces a conformational change, triggering signaling via G proteins and modulating downstream effectors such as adenylate cyclase (cAMP). PTH1R couples to G(s) G alpha proteins, mediating adenylate cyclase activation. Compared to PTH, PTHLH dissociates more rapidly from PTH1R, resulting in a faster decay of the cAMP response induced by PTHLH.
