Prostatic Acid Phosphatase (PAP) is a non-specific tyrosine phosphatase that dephosphorylates various substrates under acidic conditions (pH 4-6), including alkyl, aryl, and acyl orthophosphate monoesters and phosphorylated proteins. It exhibits lipid phosphatase activity, inactivating lysophosphatidic acid in seminal plasma, and acts as a tumor suppressor in prostate cancer by dephosphorylating ERBB2 and deactivating MAPK-mediated signaling. Additionally, it possesses ecto-5'-nucleotidase activity in dorsal root ganglion (DRG) neurons, generating adenosine from AMP to suppress pain (by similar). In microbial infection contexts, PAP forms amyloid beta-sheet fibrils (termed SEVI) in semen, which capture HIV virions, attach them to target cells, and enhance infection. These SEVI fibrils are degraded by the green tea polyphenol epigallocatechin-3-gallate (EGCG), while surfen inhibits target cell attachment and HIV infection enhancement. Similarly, PAP also boosts infection by xenotropic murine leukemia virus-related virus (XMRV).
