PP5 is a serine/threonine-protein phosphatase that dephosphorylates a wide range of proteins involved in various signaling pathways, including kinases CSNK1E, ASK1/MAP3K5, PRKDC, and RAF1, nuclear receptors NR3C1, PPARG, ESR1, and ESR2, SMAD proteins, and TAU/MAPT. It plays a role in diverse cellular processes such as apoptosis, differentiation, DNA damage response, cell survival, and regulation of ion channels or circadian rhythms, responding to steroid and thyroid hormones, calcium, fatty acids, TGF-beta, and oxidative or genotoxic stress. PP5 participates in DNA damage repair mechanisms by regulating ATM/ATR signaling and dephosphorylating PRKDC and TP53BP1. It inhibits ASK1/MAP3K5-mediated apoptosis induced by oxidative stress and positively contributes to adipogenesis by dephosphorylating and activating PPARG's transactivation function (by similar). Additionally, it dephosphorylates and counteracts the anti-adipogenic effect of NR3C1 (by similar). PP5 regulates circadian rhythms by dephosphorylating and activating CSNK1E and may modulate the TGF-beta pathway by controlling SMAD3 phosphorylation and expression levels. It also dephosphorylates TAU/MAPT, potentially regulating its function, and influences ion channels like KCNH2 through dephosphorylation (by similar). Furthermore, PP5 dephosphorylates FNIP1, disrupting its interaction with HSP90AA1/Hsp90.
