Background

PP2A alpha/beta is the catalytic subunit of protein phosphatase 2A (PP2A), a serine/threonine phosphatase that regulates a wide range of enzymes, signaling pathways, and cellular processes. As the major phosphatase for microtubule-associated proteins (MAPs), PP2A modulates the activity of phosphorylase B kinase, casein kinase 2, mitogen-stimulated S6 kinase, and MAP-2 kinase. During meiosis I in oocytes, it cooperates with SGO2 to protect centromeric cohesin from separase-mediated cleavage (by similar). PP2A dephosphorylates multiple substrates including SV40 large T antigen, AXIN1, p53/TP53, PIM3, and WEE1, while activating RAF1 through dephosphorylation at Ser-259 (PubMed references). By stabilizing WEE1 via dephosphorylation, it enforces the G2/M checkpoint. Interaction with AMBRA1 enhances PP2A's binding to MYC or FOXO3, promoting MYC degradation or FOXO3 stabilization, respectively (PubMed references). It also dephosphorylates the pyrin domain of NLRP3 to facilitate inflammasome assembly (by similar) and, together with RACK1, inhibits AKT-mTOR signaling by dephosphorylating AKT1 at Ser-473 (by similar). As part of STRIPAK complexes, PP2A regulates Hippo, MAPK, nuclear receptor, and cytoskeleton remodeling pathways (PubMed references). Within the INTAC complex, PP2A terminates unfavorable transcription elongation by dephosphorylating Pol II's CTD and SUPT5H/SPT5 (PubMed references).
