Background

PP1α is a protein phosphatase that associates with over 200 regulatory proteins to form highly specific holoenzymes, dephosphorylating hundreds of biological targets. It is essential for cell division, transcription elongation, and participates in regulating glycogen metabolism, muscle contractility, and protein synthesis. PP1α is involved in modulating ionic conductances and long-term synaptic plasticity and may play a key role in dephosphorylating substrates such as postsynaptic density-associated Ca2+/calmodulin-dependent protein kinase II. As the catalytic component of the PNUTS-PP1 complex, it promotes RNA polymerase II transcription pause-release by dephosphorylating transcription-related proteins (e.g., AFF4, CDK9) and regulates transcription termination via SUPT5H dephosphorylation. The complex also mediates POLR2A dephosphorylation during replication stress, leading to RNA polymerase II degradation. PP1α controls NEK2 kinase activity and centrosome splitting and contributes to neural tube/optic fissure closure and enteric neural crest cell migration during development (by similarity). It cooperates with CSNK1D/E to regulate PER1/2 phosphorylation rhythms for circadian period determination and inactivates FOXP3 in rheumatoid arthritis Treg cells by dephosphorylating 'Ser-418'. PP1α inhibits autophagy via ATG16L1 dephosphorylation and activates IFIH1/MDA5 and RIG-I with PPP1CC for innate immunity. As part of the SHOC2-MRAS-PP1c (SMP) holophosphatase complex, it stimulates MAPK pathway activation by dephosphorylating inhibitory sites on RAF1, BRAF, and ARAF kinases. Additionally, PP1α is required for alphavirus replication.
