PP1C gamma Antibody (YA5210) is a mouse-derived and non-conjugated monoclonal antibody, targeting to PP1C. It can be applicated for WB assays, in the background of human.

Background

PP1C is a protein phosphatase that associates with over 200 regulatory proteins to form highly specific holoenzymes, dephosphorylating hundreds of biological targets. It is essential for cell division and participates in regulating glycogen metabolism, muscle contractility, and protein synthesis. PP1C dephosphorylates RPS6KB1 and plays a role in regulating ionic conductances and long-term synaptic plasticity. It may also be important for dephosphorylating substrates such as the postsynaptic density-associated Ca2+/calmodulin-dependent protein kinase II. PP1C is a component of the PTW/PP1 phosphatase complex, which regulates chromatin structure and cell cycle progression during the transition from mitosis to interphase. In balance with CSNK1D and CSNK1E, it determines the circadian period length by regulating the speed and rhythmicity of PER1 and PER2 phosphorylation (By similarity). It may also dephosphorylate CSNK1D and CSNK1E (By similarity). PP1C regulates the recruitment of the SKA complex to kinetochores and dephosphorylates the Ser-418 residue of FOXP3 in regulatory T-cells (Treg) from rheumatoid arthritis patients, inactivating FOXP3 and impairing Treg function. Together with PPP1CA (PP1-alpha subunit), PP1C dephosphorylates IFIH1/MDA5 and RIG-I, activating them and promoting innate immune responses. PP1C is a core component of the SHOC2-MRAS-PP1c (SMP) holophosphatase complex, which regulates MAPK pathway activation. The SMP complex specifically dephosphorylates the inhibitory phosphorylation sites Ser-259 of RAF1, Ser-365 of BRAF, and Ser-214 of ARAF, stimulating their kinase activities. PP1C dephosphorylates MKI67 at the onset of anaphase, and the SMP complex enhances PP1c's dephosphorylation activity and substrate specificity.
