Description

PMS2 Antibody (YA4088) is a Mouse-derived and non-conjugated IgG2b monoclonal antibody, targeting to PMS2.

Background

PMS2 is a component of the post-replicative DNA mismatch repair system (MMR). It heterodimerizes with MLH1 to form MutL alpha, which participates in the initiation of DNA repair. MutS alpha (MSH2-MSH6) or MutS beta (MSH2-MSH3) first binds to the dsDNA mismatch, followed by recruitment of MutL alpha to the heteroduplex. In the presence of RFC and PCNA, assembly of the MutL-MutS-heteroduplex ternary complex is sufficient to activate the endonuclease activity of PMS2, introducing single-strand breaks near the mismatch to create new entry points for exonuclease EXO1 to degrade the mismatch-containing strand. DNA methylation prevents cleavage, ensuring that only the newly mutated DNA strand is corrected. MutL alpha (MLH1-PMS2) also physically interacts with the clamp loader subunits of DNA polymerase III, suggesting a role in recruiting DNA polymerase III to the MMR site. Additionally, PMS2 is implicated in DNA damage signaling, a process that induces cell cycle arrest and can lead to apoptosis in cases of severe DNA damage. PMS2 possesses ATPase activity, but ATP hydrolysis may not be necessary for proficient mismatch repair in the absence of gross structural changes.
