PKC epsilon Antibody (YA127) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to PKC epsilon.

PKC epsilon is a calcium-independent, phospholipid- and diacylglycerol (DAG)-dependent serine/threonine-protein kinase that regulates diverse cellular processes, including cytoskeletal dynamics (cell adhesion, motility, migration, and cell cycle), neuronal growth, ion channel modulation, immune responses, cancer invasion, and apoptosis. It mediates integrin-dependent cell-matrix adhesion (e.g., angiotensin-2-induced ITGB1 activation in cardiac fibroblasts) and phosphorylates MARCKS to activate PTK2/FAK, promoting cardiomyocyte spreading. In mesenchymal cells, it controls ITGB1 transport by phosphorylating vimentin (VIM), while in epithelial cells, it phosphorylates KRT8 to regulate desmosomal adhesion. PKC epsilon facilitates cell migration via IQGAP1-CDC42 signaling and is critical for HGF-induced wound healing in corneal epithelium. During cytokinesis, it forms a complex with YWHAB to regulate daughter cell separation and abscission, potentially via RHOA. In cardiomyocytes, it modulates myofilament function and excitation-contraction coupling. Additionally, PKC epsilon contributes to neurite outgrowth (via RHOA inhibition and CDC42 activation), synaptic plasticity (e.g., GABRG2 and TRPV1 phosphorylation), and prostate cancer invasion (via STAT3 activation). Downstream of TLR4, it phosphorylates TICAM2/TRAM to activate IRF3 and cytokine production. It also protects erythroid progenitors from TRAIL-induced apoptosis via BCL2 and may regulate insulin-induced AKT1 activation. by similar: PKC epsilon phosphorylates NLRP5/MATER and LRRK1, modulating AKT signaling and RAB-dependent intracellular trafficking, respectively.

  Western blot analysis of extracts from Jurkat (lane 1) and CaCO2 (lane 2) using PKC epsilon antibody. Proteins were transferred to a PVDF membrane and blocked with 5% nonfat powdered milk in PBST for 2 hour at room temperature. The primary antibody (1/1000) and loading control antibody (GAPDH, 1/3000) was diluted with 5% nonfat powdered milk in PBST at 4°C overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (1/8,000) was incubated for 45min at room temperature.
