SMAD1, a versatile transcriptional modulator, plays pivotal roles in various cellular processes, including embryonic development, cell differentiation, and tissue homeostasis. Upon binding of BMP ligands to their receptors, SMAD1 undergoes phosphorylation by activated type I BMP receptors, forming a heteromeric complex with SMAD4 that translocates into the nucleus, functioning as a transcription factor. This hetero-trimeric complex recognizes cis-regulatory elements containing Smad Binding Elements (SBEs), thus modulating the signaling network's outcome. Notably, the SMAD1/OAZ1/PSMB4 complex participates in the degradation of the CREBBP/EP300 repressor SNIP1 and positively regulates BMP4-induced expression of the odontogenic development regulator MSX1.
