PDGF Receptor beta is a tyrosine-protein kinase that functions as a cell-surface receptor for homodimeric PDGFB and PDGFD, as well as heterodimers formed by PDGFA and PDGFB. It plays essential roles in embryonic development, cell proliferation, survival, differentiation, chemotaxis, and migration. Critical for blood vessel development, it promotes the proliferation, migration, and recruitment of pericytes and smooth muscle cells to endothelial cells. It also contributes to vascular smooth muscle cell migration and neointima formation at injury sites, and is required for normal cardiovascular system development, pericyte (mesangial cell) recruitment in kidney glomeruli, and glomerular capillary network formation. The receptor induces actin cytoskeleton rearrangement and membrane ruffle formation. Ligand binding activates multiple signaling cascades, phosphorylating PLCG1, PIK3R1, PTPN11, RASA1/GAP, CBL, SHC1, and NCK1, thereby regulating downstream effectors such as diacylglycerol, inositol 1,4,5-trisphosphate, Ca²⁺ mobilization, protein kinase C, AKT1, HRAS, RAF1, and MAPK1/ERK2/MAPK3/ERK1.
