Background

Lamin A is an intermediate filament protein that, along with Lamin C (by similar), forms the major components of the nuclear lamina, a fibrous meshwork on the nucleoplasmic side of the inner nuclear membrane. It provides structural support by bridging the nuclear envelope and chromatin, playing pivotal roles in nuclear assembly, chromatin organization, nuclear membrane dynamics, and telomere maintenance. Lamin A/C regulates nuclear mechanical properties to influence matrix stiffness, with its structural integrity tightly controlled during cell cycle progression (e.g., disassembly in prophase and reassembly in telophase). Mammalian lamina contains equal amounts of Lamin A and C. The protein participates in DNA repair by being recruited to double-strand breaks by XRCC4 and IFFO1 to immobilize broken DNA ends, preventing chromosomal translocations. It is essential for peripheral nervous system development, skeletal muscle formation, muscle satellite cell proliferation (by similar: osteoblastogenesis and bone formation while preventing fat infiltration into muscle/bone marrow to maintain tissue integrity). Cardiac homeostasis requires Lamin A/C, though prelamin-A/C may promote vascular smooth muscle cell senescence via mitotic disruption and DNA damage-induced genomic instability.
