Phospho-FOXO3A(Ser253) Antibody (YA196) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to Phospho-FOXO3A(Ser253).
Background

FOXO3a is a transcriptional activator that recognizes and binds to the DNA sequence 5'-[AG]TAAA[TC]A-3', regulating processes such as apoptosis and autophagy. In skeletal muscle, it acts as a positive regulator of autophagy: under starvation conditions, FOXO3a enters the nucleus after dephosphorylation and binds to the promoters of autophagy-related genes (e.g., GABARAP1L, MAP1LC3B, and ATG12), activating their expression and leading to proteolysis of muscle proteins (by similar). It triggers apoptosis in the absence of survival factors, including neuronal cell death under oxidative stress. Additionally, upon phosphorylation by MAPKAPK5, FOXO3a promotes the expression of miR-34b and miR-34c, post-transcriptional regulators of MYC. During metabolic stress, it translocates to mitochondria to enhance mtDNA transcription. FOXO3a also regulates chondrogenic commitment of skeletal progenitor cells by activating SOX9 expression under low lipid conditions (by similar) and suppresses fatty acid oxidation. Moreover, it plays a key role in regulatory T-cell (Treg) differentiation by activating FOXP3 expression.
