eIF4G is a component of the eIF4F protein complex, which recognizes the mRNA cap structure, unwinds 5'-terminal secondary structures in an ATP-dependent manner, and recruits mRNA to the ribosome. It exists in two mutually exclusive complexes, either with EIF1 or EIF4E. In complex with EIF1, eIF4G facilitates leaky scanning, particularly to bypass cap-proximal start codons. When bound to EIF4E, it counteracts EIF1-EIF4G1-mediated scanning and directly locates the start codon via a TISU element without scanning.
